Lipid nanoparticles(LNPs) are typically composed of four components. (1) cationic or ionizable lipids, which are smart macromolecules that can be functionalized to improve mRNA entrapment, enhance cellular uptake efficiency, and promote endosomal escape; (2) lipid-anchored polyethylene glycol (PEG), which is used to reduce particle size, prevent particle aggregation, increase circulation time, and reduce uptake by non-target cells; (3) cholesterol that stabilizes the LNPs complex; and (4 ) phospholipids that support the lipid bilayer structure.
